The Q Exactive GC coupled with IRMS (QE GC-IRMS) is a unique tool for metabolite research, allowing for the simultaneous determination of isotopically labeled metabolites and their corresponding structural information. This approach combines the Thermo Scientific Delta V IRMS or 253 Plus IRMS system with the Thermo Scientific Q Exactive GC connected to a single Thermo Scientific Trace 1310 GC, providing a new method for metabolite research. The system enables ultra-high mass resolution, sub-ppm mass accuracy, and a large dynamic range for untargeted and targeted metabolite research. Compound-specific stable isotope analysis is used to provide insights into the metabolism of compounds, and deuterium labeling is used to identify metabolites. The QE GC-IRMS system has been used to effectively detect the deuterium label of metabolites and provide structural information of the metabolites at the same time. An elimination study with oral administration of doubly- and triply deuterated testosterone has been performed, and urine samples were collected and processed according to established sample preparation procedures. The Q Exactive GC-IRMS system was used to measure glucuronidated, sulfoconjugated, and unconjugated steroids, and the results show the ability of the system to detect and identify metabolites.
