Summary

Die vorliegende Dissertation beschäftigt sich mit der voltammetrischen Detektierung der DNA-Hybridisierung von Oligonukleotiden und PCR-Produkten an Goldelektroden mit Osmiumtetroxid als kovalentem Marker. Die Arbeit umfasst die Entwicklung und Charakterisierung eines elektrochemischen Biosensors, der die Hybridisierung von DNA-Molekülen an einer Goldelektrode mit Hilfe des Markers [OsO4(bipy)] detektiert. Die Ergebnisse zeigen, dass der Biosensor eine hohe Selektivität und Signalstabilität aufweist und für die Detektierung von DNA-Hybridisierungen geeignet ist. Die Arbeit behandelt auch die Kinetik der Markierungsreaktion und die Einflüsse von Temperatur und Hybridisierungsdauer auf die Detektierung. Der Ausblick diskutiert mögliche Anwendungen des Biosensors, wie z.B. DNA-Chips, Markierung von Reportersträngen und RNA sowie die Verwendung von Aptameren und anderen Markern.
