Summary

The advent of next-generation sequencing has revolutionized the sequencing and availability of whole genome data for numerous plant species. However, the genome sequencing of major staple food crops, such as wheat, barley, and rye, has been challenging due to their complex polyploid nature and large genome size. Recent advancements in sequencing technology have enabled the sequencing of wild grass relatives, such as Brachypodium, which has provided a framework for deciphering the complex genomes of cereal crops. The sequencing of barley and rye has also been accomplished, with barley's 5 GB genome sequenced in 2012 and rye's 8 Gb genome partially sequenced using chromosome survey sequencing and high-throughput transcript mapping. Wheat, with its gigantic allohexaploid genome, remains a significant challenge, but efforts have been made to sequence its non-repetitive content to a 5X coverage.
