Summary

Apoptolidin is a selective cytotoxic agent that inhibits F0F1-ATPase, a mitochondrial protein. It was identified as a macrolide that selectively kills transformed rat glial cells while sparing untransformed cells. The mechanism of action of apoptolidin involves the inhibition of mitochondrial F0F1-ATP synthase, which is similar to other macrolides such as oligomycin. Apoptolidin-induced cell death is independent of p53 status, inhibited by BCL-2, and dependent on the action of caspase-9. The selectivity of apoptolidin correlates with that of other known anti-fungal natural products that inhibit the eukaryotic mitochondrial F0F1-ATP synthase.
