Summary

Oncogenic RAS promotes leukemic transformation of CUX1-deficient cells. CUX1 mutations are prevalent in myeloid neoplasms and often occur early in disease pathogenesis, carrying a poor prognosis. Combining CUX1 deficiency with oncogenic RAS mutations leads to the development of acute myeloid leukemia (AML) in mice, whereas either mutation alone does not. Oncogenic RAS increases self-renewal of CUX1-deficient hematopoietic stem/progenitor cells, and CUX1 knockdown amplifies RAS signaling. Double mutant cells are responsive to PIK3 or MEK inhibition, suggesting a potential therapy for malignancies with CUX1 inactivation.
