Summary

Light-sheet fluorescence microscopy enables rapid 3D imaging of biological samples with high contrast and minimal sample exposure. To achieve a wide field-of-view without compromising axial resolution, propagation-invariant light-fields like Bessel and Airy beams are used. However, these beams have a broad transversal structure, and the Airy beam's fine structure is suppressed in two-photon excitation. A symmetric and planar Airy light-sheet is demonstrated for two-photon excitation, which does not rely on deconvolution and enables high isotropic resolution across the entire field-of-view, as shown in imaging a 0.6mm-thick volume of Wistar rat brain.
