Summary

Valtcheva et al. injected viral tracers into oxytocin neurons of maternal mice to label brain areas sending auditory information to these neurons. They found that areas processing conscious auditory responses are not directly connected to oxytocin neurons, but instead send information through the posterior thalamus, specifically the posterior intralaminar nucleus (PIL). The PIL, part of the non-lemniscal auditory pathway, mediates unconscious perception of sounds and projects to the oxytocin neurons, influencing maternal care. Silencing PIL signaling reduced mothers' willingness to protect their pups, and oxytocin release in the ventral tegmental area (VTA) was negligible when PIL signaling was silenced. The study provides a mechanistic understanding of how sensory cues from offspring activate oxytocin release and promote maternal care, raising questions for future research on the sufficiency of this pathway to drive maternal behavior and its potential alteration in postnatal conditions like postpartum depression.
