The bacterial flagellar motor is powered by an electrochemical potential difference of ions across the cell membrane, generating torque through rotor-stator interactions and proton translocation. The stator complex, composed of MotA and MotB, dynamically assembles and disassembles from the motor, with the peptidoglycan-binding (PGB) domain of MotB anchoring the complex to the peptidoglycan layer. The crystal structure of the periplasmic region of MotB reveals a dimeric structure with a PGB domain, and mutation analyses indicate that dimerization is crucial for motor function. A proposed mechanism suggests that the MotA/B complex diffuses in the cytoplasmic membrane, and upon incorporation into the motor, undergoes a conformational change to anchor the PGB domain to the peptidoglycan layer and open the proton channel, activating the motor.
