Summary

The interaction of polylysine with DNA can be divided into primary and secondary interactions, with most studies focusing on the aggregated phase. This study used fluorescence depolarization to measure the primary interaction, finding it to be independent of aggregation and factors affecting it. The results showed that the interaction is irreversible, with the fraction of polylysine bound depending on the path taken to reach a certain ionic strength. pH studies indicated pK shifts of the chromophore due to charge effects of the DNA phosphates, and turbidity was found to be sensitive to experimental parameters and not a quantitative measure of the complex.
