En France, 350 000 nouveaux cas de cancer sont identifiés chaque année, liés à des dysfonctionnements cellulaires dus à des mutations. Les perturbations du génome favorisent le développement de maladies comme le cancer. Le développement d'un cancer se déroule en plusieurs étapes : prolifération anormale de cellules, formation d'une tumeur, atteinte des tissus voisins et métastases. Les cellules cancéreuses acquièrent des capacités de prolifération accrues suite à des mutations, notamment sur le gène p53, qui joue un rôle anti-mutagène. Les cancers peuvent être favorisés par des agents mutagènes tels que le tabagisme, l'amiante et les rayons UV, ainsi que par des virus comme le papillomavirus. La lutte contre les cancers repose sur la connaissance de leurs causes, le dépistage, la vaccination et l'hygiène de vie.
