Summary

A voxel-based meta-analysis of grey-matter volume studies in major depression (MDD) and bipolar disorder (BD) identified common and distinct volumetric alterations. The analysis included 50 MDD datasets (4101 individuals) and 36 BD datasets (2407 individuals). Conjunction analysis revealed both disorders showed reduced grey-matter volume in clusters of the dorsomedial and ventromedial prefrontal cortex, anterior cingulate cortex, and bilateral insula. Group comparisons indicated MDD exhibited more substantial reductions in the right dorsolateral prefrontal cortex, left hippocampus, cerebellum, temporal regions, and parietal regions compared to BD. These findings demonstrate that MDD and BD share specific grey-matter volume reductions while also displaying distinct patterns, suggesting this combination of similarities and differences could inform diagnostic biomarker development for these conditions.
