Summary

The stomach and colon have a two-layered mucus structure, with an inner attached layer and an outer less dense unattached layer. Mucins, glycosylated proteins, are crucial for gastrointestinal luminal protection. The inner mucus layer is bacteria-resistant, while the outer layer serves as a home for commensal microorganisms. Disruption of the mucus layer can lead to intestinal diseases such as ulcerative colitis. A human-relevant in vitro model of colon mucus physiology has been developed using organ-on-a-chip and organoid technologies, allowing for noninvasive visualization of mucus accumulation and simulation of mucus bilayer creation. The model can be used to investigate the role of mucus in human intestinal homeostasis and diseases.
