Summary

The aim is to investigate the permeation of hydrocortisone across the PermeaPad Barrier in a side-by-side set-up. Hydrocortisone solution is prepared and added to the donor compartment, while PBS is added to the acceptor compartment. Samples are taken every 30 minutes for 3.5 hours and quantified using methods such as UV spectroscopy or HPLC-UV. The apparent permeability (Papp) is calculated by plotting the cumulative amount of hydrocortisone permeated against time and normalizing by the donor start concentration. The PermeaPad Barrier is considered to have correct permeation properties when the Papp is 15 ± 1.5 × 10-6 cm/s.
