Summary

Affinity maturation is a key protein engineering technique used to improve antibody affinity and binding interactions in vitro. Various display technologies and maturation methods have been developed, but limitations in display capacity remain a challenge. An unbiased approach using DNA recombination and shuffling techniques, combined with ribosome display, was used to affinity mature a difficult antibody lineage, resulting in a 50-fold improvement in affinity and potency. The approach involved recombining mutations from all six complementarity-determining regions, creating fresh combinatorial diversity and producing antibody variants with mutations spanning the length of the antibody construct. The results highlight the importance of exploring a larger sequence space and the potential for unpredictable improvements in affinity and binding interactions.
