Summary

Nitrocefin is used to detect β-lactamases through chromogenic assays. A stock solution (10 mg/ml) is prepared in DMSO and then diluted to a working solution (0.5-1.0 mg/ml) in PBS. The working solution appears yellow and turns red upon exposure to β-lactamase. Various detection assays include slide surface, direct contact, broth suspension, lysed cell, and filter paper assays, all producing a positive result within 20-30 minutes. Spectrophotometric analysis measures β-lactamase activity based on Nitrocefin degradation.
