Summary

CD spectroscopy is used for structural analysis of proteins, with CD spectra usually observed from UV region below 260 nm to the vacuum ultraviolet region, but water absorption in the vacuum UV region can interfere. The J-1500 CD spectrometer allows measurement with high signal to noise ratio in the vacuum UV region. CD measurement of protein water solution was performed down to the vacuum UV region using the J-1500 CD spectrometer and multivariate SSE program, with results showing high S/N ratio down to 174 nm. Secondary structure estimation was carried out using the multivariate SSE program, with improved estimation of β-sheet structure achieved by applying PLS/PCR multivariate analysis method.
