Summary

The term "allosteric" refers to a mechanism where a protein can exist in two or more distinct conformations that differ in their affinity for a ligand, with the binding of an allosteric regulator causing a conformational change that alters the protein's activity. The original definition by Monod et al. (1965) limited the term to oligomeric molecules showing cooperativity, but it is now commonly used to describe monomeric receptors as well. In the context of agonist activation of an ion channel, the term "allosteric" can be applied if the agonist has a higher affinity for the open channel than for the shut channel, causing a shift in the equilibrium towards the open channel.
