Summary

Dendritic cells have a cell-intrinsic sensor for HIV-1 that activates antiviral innate immunity, inducing dendritic cell maturation, a type I interferon response, and T-cell activation when the cells are productively infected with HIV-1. This response is dependent on the interaction between newly synthesized HIV-1 capsid and cellular cyclophilin A, and the subsequent activation of the transcription factor IRF3. The sensor is not typically engaged due to dendritic cell resistance to HIV-1 infection, but its activation may be necessary for generating an effective HIV-1 vaccine.
