Accurate evaluation of a new compound's pharmacokinetic (PK) and pharmacodynamic (PD) profile is essential in the drug development process. Preclinical PK evaluation in animals characterizes the relationship between drug dose, route of administration, and blood concentrations. The PD profile determines the relationship between tissue or fluid concentration and therapeutic efficacy and side effects. Factors affecting PK profiles and behavioral manifestations include route of administration, dose, and stress. Stress can alter drug bioavailability and motor response, and common laboratory procedures can cause significant alterations in stress indicators. The method of drug administration is critical, and automated dosing may be less stressful than manual dosing. A study compared automated and manual oral dosing on nicotine absorption, conversion, and locomotor-activating effects in rats, finding that nicotine absorption was more rapid with automated dosing, but did not significantly alter conversion to cotinine or motor activity. The results suggest that the method of dosing can influence pharmacokinetics and should be considered in study designs.
