Summary

Viruses are masters of camouflage and deception, exploiting cells and organisms to disseminate. To enter a host cell, a virus must deliver its genome and accessory proteins, often by manipulating the host cell's machinery. However, the process of capsid disassembly, or "uncoating," remains poorly understood. Researchers have developed an in vitro single-molecule fluorescence imaging assay to study the uncoating kinetics of individual HIV capsids, allowing them to classify virions into three categories based on their uncoating behavior and define two discrete uncoating events: "capsid opening" and "lattice disassembly." This approach enables the study of the effects of individual host molecules and drugs on the uncoating process, providing new insights into the mechanisms of viral infection and potential targets for antiviral therapies.
