Summary

A reversed-phase high-performance liquid chromatography (RP-HPLC) method was developed and validated for the quantification of mangiferin, a polyphenol xanthone glycoside, in Mangifera indica. The method used isocratic elution with 0.1% formic acid:acetonitrile (87:13) as the mobile phase, a flow rate of 1.5 ml/min, and detection at 256 nm. The method was validated for linearity, precision, accuracy, limit of detection, limit of quantification, and robustness, showing good linearity, high reliability, and reproducibility. The recovery values were 100.47, 100.89, and 100.99 at three different levels, and the results remained unaffected by small variations in analytical parameters. Liquid chromatography-mass spectrometry confirmed the presence of mangiferin with an M/Z value of 421. The developed assay is simple, rapid, and reliable for the determination of mangiferin from M. indica.
