Eurhopalothrix reichenspergeri, an ant species, has undergone taxonomic modifications, with its valid species status recently supported. This study provides an addendum to the last taxonomic study, exploring immature stages, spermatozoa morphology, and cytogenetics. A colony was collected in an Atlantic Forest fragment in Brazil, and species identification was confirmed. The study includes high-resolution images of mature stages, available in AntWeb, and specimens deposited in the Coleção Entomológica do Laboratório de Coleoptera.

Larval external morphology was described, with the larva being long, stout, and hairy, approximately 2.4 mm in length. The head is large and ventrally directed, with a cranium almost twice as wide as high. Mandibles are strongly sclerotized, long, and slender, with a clearly developed apical tooth. Maxilla and labium are described in detail, with the maxilla bearing a palpus with three visible divisions and the labium having three visible divisions on its palpus.

Spermatozoa morphology was analyzed, with total and nucleus lengths measured. Mitotic metaphase chromosomes were obtained, and female and male karyotypes were analyzed. The results show that the female karyotype has a specific number and chromosomal morphology, with chromosomes classified as metacentric, submetacentric, subtelocentric, and acrocentric. Fluorescent in situ hybridization (FISH) experiments were conducted to map clusters of 18S ribosomal DNA. The study provides new information on the morphology, cytogenetics, and spermatozoa of Eurhopalothrix reichenspergeri, increasing knowledge of this species.
