Summary

Reactive oxygen species (ROS) are by-products of mitochondrial metabolism involved in cellular signaling pathways and tissue injury. The mitochondrial respiratory chain is a major source of ROS, but how ROS release depends on oxygen concentration is debated. This study measured the rate of H2O2 release by intact brain mitochondria using an Amplex UltraRed assay and a high-resolution respirometer. The results show that H2O2 release is linearly dependent on oxygen concentration, with the highest rate occurring in conditions of reverse electron transfer when mitochondria oxidize succinate or glycerol 3-phosphate. Complex II can contribute to ROS generation even in the absence of its substrate succinate when the quinone pool is reduced by glycerol 3-phosphate. The study demonstrates that ROS generation in brain mitochondria is lower under hypoxic conditions than in normoxia, and highlights the importance of reverse electron transfer in the brain, particularly during ischemia/reperfusion.
