Summary

- Purpose: To analyze sequence variances and phylogenetic relationships among seven River Nile teleostei species using partial mitochondrial cytochrome-b gene sequences.
- Species Studied: Schilbe mystus, Barbus bynni, Oreochromis niloticus, Bagrus bajad, Mormyrus kannume, Lates niloticus, and Clarias gariepinus.
- Methodology:
- DNA extraction from fish muscle tissues.
- PCR amplification of the mitochondrial cytochrome-b gene using specific primers.
- Sequencing of PCR products and alignment with reference sequences.
- Phylogenetic analysis using Maximum Likelihood (ML) and Unweighted Pair Group Method with Arithmetic Mean (UPGMA) methods.
- Findings:
- A three-nucleotide deletion in Siluriformes catfish sequences.
- Similar nucleotide composition to actinopterygian fishes.
- High variation at the nucleic level, supporting the gene's suitability as a DNA marker.
- Close genetic relationship among teleost actinopterygian River Nile fish species.
- Close genetic and evolutionary relation of early divergent orders: Acipenseriforms (Sturgeons) and Semionotiformes (Alligator gars).
- Implications: New sequence data could contribute to successful fish management, conservancy, and aquaculture objectives. Additional DNA sequences from other River Nile teleost family representatives could enhance future fish molecular phylogeny studies.

Description

The PCR products from mitochondrial Cytochrome-b gene were sequenced for seven River Nile teleost species. A three-nucleotides cytochrome-b gene deletion existed in Siluformes catfish sequences, possibly accounting for species adaptation and monophyletic evolution. Analyzed sequences showed similar nucleotide composition to actinopterygian fishes.
