Summary

Drug resistance in cancer is often caused by genetic mutations, which can occur randomly or be induced by drugs. Researchers have developed mathematical models to study drug resistance, including the Goldie and Coldman model and a model by Iwasa et al. that estimates the probability of developing resistance by the time a tumor is detected. However, these models assume a homogeneous population of cancer cells, whereas recent evidence suggests that tumors are heterogeneous and maintained by a small subset of stem cells. A new model incorporates the heterogeneity of the population and estimates the probability of developing resistance, taking into account the modes of division of stem cells, including asymmetric division and symmetric renewal. The model is applied to clinical data from a study on chronic myelogenous leukemia, suggesting that leukemia stem cells tend to renew symmetrically rather than asymmetrically.
