Summary

The pyriproxyfen metabolite, 4′–OH–PPF, disrupts thyroid hormone signaling in neural stem cells, modifying neurodevelopmental genes affected by ZIKA virus infection. Researchers found that 4′–OH–PPF decreased thyroid hormone signaling and increased Musashi-1 expression, a protein necessary for ZIKV replication, in neural stem cells. Co-exposure to 4′–OH–PPF and ZIKV decreased viral replication but also dysregulated key genes in thyroid hormone signaling and neuroglial commitment, suggesting a synergistic block of transcriptional machinery. The study concludes that 4′–OH–PPF is a thyroid hormone antagonist that alters neural stem cell processes, potentially aggravating the microcephaly phenotype when combined with ZIKV infection.
