Summary

Förster resonance energy transfer (FRET) is a phenomenon of non-radiative transfer of optical excitation from a donor to an acceptor molecule, used to measure separating distance between molecules. Lanthanides like Europium Cryptate have long emission lifetimes and high transfer efficiency to acceptors like Allophycocyanin. Time-Resolved FRET (TR-FRET) measures donor and acceptor intensities in "gated" mode, but has drawbacks like emission spectrum overlap and requirement of two detectors. A time-resolved fluorescence format can increase sensitivity and reduce cost by applying multi-channel scaling mode to measure donor emission time-responses, allowing separation of microsecond background and avoiding spectral "cross-talk".
