Summary

The structure and transcriptional activity of chromatin from logarithmically growing yeast cells and cells in the stationary phase were compared. Both chromatins showed similar nucleosomal repeat patterns, but growing phase chromatin was more susceptible to DNase I digestion and had 5-20 times more endogenous transcription activity. The transcriptional differences were due to differences in in vivo initiation. A rapid technique for isolating yeast nuclei was developed, and transcription initiation in isolated nuclei was studied using nucleoside 5'-[y-S] triphosphates as affinity probes. Endogenous RNA polymerase III initiated 5S and pre-tRNA transcription in vitro, as well as a discrete distribution of RNA species up to 28S.
