This study addresses the threat facing medicinal plants in Jordan due to habitat perturbation, overharvesting, climate change, and competition from invasive species. It focuses on two rare and endangered Jordanian plants: Achillea millefolium (Jerash) and Moringa peregrina. The primary objectives are to establish an efficient in vitro micropropagation method for these plants and assess their antimicrobial activity.

Methods:

1. Seed Disinfection and Germination: Seeds were surface sterilized, germinated on agar water media, and then transferred to MS medium with 3% sucrose for shoot culture.

2. In Vitro Shoot Proliferation: Microshoots were cultured on MS medium with different concentrations of 6-benzyl amino purine (BAP) to optimize growth. After four weeks, the number of shoots per explant and microshoots was recorded.

3. Antimicrobial Activity Assessment: Both in vitro (plantlets) and ex vitro (field leaf plants) extracts were evaluated against various bacteria using ethanolic extracts.

Results:

Achillea millefolium and Moringa peregrina successfully multiplied in vitro, with notable antimicrobial activity against tested microorganisms. The study highlights the potential of these two plants as sources of medicinal compounds and their promise for conservation through in vitro propagation techniques.
