Sensitivity and specificity improve when nucleoids are incubated with bacterial repair endonucleases that convert specific lesions (e.g., UV-induced pyrimidine dimers) to breaks. DNA repair kinetics can be monitored by measuring residual damage after exposure to damaging agents or by assessing repair activity in cell extracts. While alkaline conditions (pH >13) enhance tail visibility and detect a wider range of damage, high pH is not essential for sensitivity; the assay's simplicity, speed, and cost-effectiveness make it widely used despite misconceptions about its mechanism. The comet tail represents relaxed DNA loops pulled by electrophoresis, not a direct indicator of break quantity alone.
