Summary

Thiol-modified oligonucleotides require deprotection prior to use by cleaving the disulfide bond to generate free thiol groups. Two protocols are provided: one using DTT (Dithiothreitol) and the other using TCEP (Tris(2-carboxyethyl)phosphine). The DTT protocol involves incubating the oligonucleotide with 100 mM DTT at room temperature for 30 minutes, while the TCEP protocol involves incubating with 0.1 M TCEP for 1 hour. After reduction, excess reagent can be removed through methods like ethanol precipitation, spin column purification, or dialysis. The deprotected oligonucleotides should be stored under inert atmosphere or in a solution containing 10 mM DTT or TCEP to prevent reoxidation.
