Summary

The study investigated κ-opioid receptor expression in cell cultures and human brain, aiming to develop new opioid receptor-specific ligands and examine receptor expression in the nervous system. The research focused on four main areas: developing stereoselective opioid receptor-specific ligands, studying κ-opioid receptor expression during development, investigating regulation of κ-receptor expression by chronic opioid signals, and comparing cellular localization of κ-opioid receptor protein and mRNA in human brain cortical sections and rat cerebellar granule cell cultures. The results included the localization of κ1-opioid receptor mRNA in the human frontal cortex area 10 and the detection of specific hybridization signals in pyramidal-like neurons and type II astrocytes. The study concluded that κ1- and κ2-opioid receptors could be expressed by the same cells and that the sequences of κ1- and κ2-opioid receptor mRNAs are highly homologous or identical in the region recognized by the oligonucleotide probe.
