Summary

Carrot hypocotyl seedlings were transformed with Agrobacterium tumefaciens containing the CaMV 35S promoter, β-glucuronidase coding sequence, and nopaline synthase polyadenylation sequences. Kanamycin-resistant calli were obtained in 5.8% of explants after 6 weeks, and Southern blot analysis confirmed transformation with 1-3 copies of the GUSII construction. GUS activity in transformants was 5-250 fold over background. A simple and reliable method for carrot transformation and somatic embryogenesis was reported, using freshly germinated carrot hypocotyls sliced and precultured for 2 days on medium with 2,4-D, followed by Agrobacterium co-cultivation and selection on kanamycin and carbenicillin.
