High levels of mitochondrial glycerol-3-phosphate dehydrogenase are found in pancreatic islets, 40-70 times higher than in liver, whole pancreas, heart, or skeletal muscle. The enzyme's activity is comparable to the cytosolic NAD-linked glycerol phosphate dehydrogenase, suggesting a possible glycerol phosphate shuttle in pancreatic islets. Diazoxide, an inhibitor of insulin release, inhibits the mitochondrial glycerol phosphate dehydrogenase in pancreatic islets at concentrations that also inhibit insulin release and CO2 formation from glucose. The results support the hypothesis that the mitochondrial glycerol phosphate dehydrogenase plays a key role in stimulus-secretion coupling in pancreatic β cells during glucose-induced insulin release.
