Summary

Zn-substituted cytochrome c (Zn-Cytc) was prepared and its photophysical properties investigated using single-molecule and ensemble-averaged spectroscopy. Zn-Cytc was prepared by replacing the iron ion in cytochrome c with zinc, and its absorption spectrum showed characteristic peaks at 423, 549, and 584 nm. The fluorescence intensity trajectory (FIT) of single Zn-Cytc molecules was measured using total internal reflection fluorescence microscopy (TIRFM), and the results showed that the FITs of >150 single molecules could be used to determine the on and off times of the folded and unfolded Zn-Cytc. The fluorescence lifetime of single Zn-Cytc molecules was also measured using confocal laser scanning microscopy, and the results showed that the fluorescence lifetime was affected by the presence of guanidine hydrochloride (GdHCl).
